If I have a fuel pressure regulator installed on an early Spyder, should I still run a fuel return line?

  Smitty says:  Rusty I never tried using a regulator on a turbo.  GM didn't see it necessary, and neither have I.  But that is beside the point, and not what you asked.  Some turbos are subject to vapor lock.  Since you live in a hot area of the country you might as well leave the return line there to help alleviate that problem.
